Simultaneous determination of five ingredients in Qingkailing granules by HPLC-MS/MS method

ABSTRACT  Objective: To establish a HPLC-MS/MS method for simultaneous determination of baicalin,jasminoidin,cholic acid,hyodeoxycholic acid and chlorogenic acid in Qingkailing granules.Methods: Separation was performed on an Agilent Zorbax Extend-C18 column(100 mm×2.10 mm,3.5 μm).The mobile phase was composed of acetonitrile(phase A) and 10 mmol/L ammonium acetate water solution(phase B) with gradient elution(0-1 min,5%→5% phase A,1-5 min,5%→52% phase A,5-8 min,52%→5% phase A) at a flow rate of 0.6 ml/min.Detection was performed on a triple quadrupole tandem mass spectrometer by multiple reactions monitoring(MRM) via electrospray ionization(ESI) source in negative ionization mode.Results: Method showed good linearity in baicalin,jasminoidin,cholic acid,hyodeoxycholic acid and chlorogenic acid(r>0.999 0).Mean recoveries were (98.6±2.42)%-(104.4±4.03)%.The precision,reproducibility and stability were all good.The contents of baicalin,jasminoidin,cholic acid,hyodeoxycholic acid and chlorogenic acid in the 4 batches of Qingkailing granules were 5.56-7.10,0.61-0.76,5.74-7.64,7.08-8.81,0.17-0.35 mg/g,respectively.Conclusion: The method was proved to be simple,accurate and reproducible,and could be used for quality control of Qingkailing granules.
